Immunoglobulin (IG) and T cell receptor (TR) diversity and clonotype follow

vaccination, cancer, infectious and immune diseases represent a major challenge owing to
the enormous complexity of the data generated.
The number of antigen receptors, IG or
antibodies and TR of the adaptive immune response in vertebrates with jaws, is
indeed
almost unlimited (2.10

per individual in humans). This huge diversity results from complex
mechanisms in the synthesis of the variable (V) domains, that include DNA molecular
rearrangements of the V, diversity (D) and joining (J) genes, N
-
diversit
y at the V
-
(D)
-
J
junctions and, for IG, somatic hypermutations. The specificity of the V domains is conferred
by the complementarity determining regions (CDR) and more particularly the CDR3. IMGT®,
the international ImMunoGeneTics information system® (
http
://www.imgt.org
) [1], has
developed IMGT/HighV
-
QUEST [2,3], the high throughput version of IMGT/V
-
QUEST for the
analysis of sequences obtained by Next Generation Sequencing (NGS) and deep
sequencing. IMGT/HighV
-
QUEST provides a detailed sequence analysis o
f IG or TR V
domains based on the IMGT
-
ONTOLOGY concepts.
The IMGT/HighV
-
QUEST paradigm
defines standards for genotype/haplotype analysis and characterization of IMGT clonotypes
for clonal diversity and expression and achieves a degree of resolution for ne
xt generation
sequencing verifiable by the user at the sequence level
. IMGT/HighV
-
QUEST standardized
immunoprofiling allows comparison between different B and T cell populations and immune
situations and characterization of potential therapeutic antibodies

from combinatorial
libraries.

IMGT/HighV
-
QUEST has been freely available for use for academics on the IMGT®
Home page (http://www.imgt.org) since October 2010. More than 880 million sequences
were analysed in 2013.
