2. OBJECTIVES

The present course is related to the integration of basic aspects of biotechnology with
comprehensive biochemical engineering, to give the student understanding of technological
aspects of bulk, recombinant and fine biotechnology production, including comprehensive
modes of operation in the sense of bioprocessing upstream and downstream engineering,
including aspects of scale-up and scale-down procedures from laboratory to industrial scale.

3. SYLLABUS

Microbiology in biotechnology. The microbial cell, its function and structure. Cell
metabolism. Energetics and transport. Basic aspects of gene engineering and protein
engineering. Enzymes and enzymatic reactions.
Bioprocess kinetics. Homogenous and heterogeneous reactions, order of reaction, microbial
specific growth rate, Monod kinetics. Substrates and bioprocess kinetics.
Upstream bioprocessing. Process sterility and sterilisation of liquid and gas phase. Fibrous
filters and membrane filters.
Bioprocess modes of operation. Submerged and solid state bioprocessing. Batch, fed batch
and continuous modes of operation (chemostat, pH-stat, turbistat, nutristat, auxostat).
Bioprocess instrumentation. Methods and measurements in biotechnology On-line, in-line and
off-line process control. Measurements of process temperature, pressure, power imput, foam
control, flow control, viscosity, pH, redox potential, oxygen partial pressure, substrate content
control, biomass measurements etc. Biosensors.
Solid state bioprocessing. Upstream, bioreactors, heat and mass transfer, downstream
processing. Bioprocess economics, submerged versus solid state bioprocessing.
Bioreactors for fine biotechnology products: plant and animal tissue bioreactors, fluidized bed
reactors.
Scale-up, scale-down aspects. Process integrated systems.

4. PREREQUISITES

Overview of microbiology, biochemistry, microbial physiology, chemical and biochemical
engineering as well as industrial microbiology and gene engineering.
