Clinical Cell and Vaccine Production Facility

(CVPF)
: The Clinical Cell and Vaccine
Production Facility (CVPF) facilitates the bench to bedside translation of novel
allogeneic and autologous cellular
-
based vaccines and

biological therapies as the next
generation of infused therapeutics. The CVPF provides focused scientific, technical and
regulatory support for investigator initiated investigational new drug applications (INDs)
in cell and gene therapy. Under complianc
e of FDA 21 CFR Parts 210 and 211, the
CVFP performs cell processing on a range of different cell types, to include bone marrow
derived CD4+ and CD8+ T lymphocytes, dendritic cells, and marrow stromal cells (AKA
mesenchymal stem cells). Processing include
s isolation/enrichment of particular cell
lineages, cryopreservation and storage of cells and tissues, RNA isolation from tumor,
growth and expansion of cells, and transduction (insertion) of experimental therapeutic
genes into cells. In each of these cas
es, cells are expanded ex vivo with or without
genetic modification to the cells before being reintroduced back into the patient, either
alone or in combination with other therapies such as vaccines.

Flow Cytometry and Cell Sorting Facility:

The Flow Cytometry

and Cell Sorting
Resource Laboratory (FCCSRL) is dedicated to providing Penn investigators access to
high
-
quality, cost
-
effective flow cytometric services, as well providing the scientific
expertise necessary to effectively integrate this technology into
their research projects.
New multiparametric techniques using sensitive fluorescent probes, now available in a
multitude of colors, make cell sorting and analysis (flow cytometry) a rapid, accurate and
objective method to measure multiple characteristics o
f large numbers of individual cells
simultaneously.

Human Immunology Core:

The HIC is supported by the Cancer Center Shared Grant
and the C
enter for AIDS Research. Services include:
multiplex Immunological assays
which allow the simultaneous characterization of multiple cytokines (or phosphoprotein
or mRNA); flow cytometry based immunophenotyping and proliferation (CFSE) assays;
cellular assa
ys such as B and T cell ELISpot; primary lymphocyte isolation and sub
-
population enrichment; cryopreservation and banking of PBMCs and/or plasma from
longitudinal studies for concurrent immunological analysis.

Microarray Core
: The Penn Molecular Profiling Facility provides instrumentation and
expertise for DNA and RNA profiling. Microarrays and other highly parallel technologies
provide the means for measuring the identity and abundance of DNA and RNA for
targeted genes, or th
e whole genome, in a biological sample. The Facility supports
microarrays from Affymetrix, Agilent and Illumina, as well as assays for Sequenom,
Luminex, Fluidigm and TaqMan.

Next Generation Sequencing Core
: The NGSC offers ultra high throughput sequencing
services for the PSOM research community. We offer library quality assessments,
sequencing, and optional preliminary data analysis for a wi
de variety of experimental
protocols including ChIP

seq, RNA

Seq, HITS
-
CLIP, miR
-
Seq, exome capture, and BIS
-
seq. We offer limited library preparation services, but can advise on library preparation
techniques. We have two Illumina hiSeq2000s for large

sca
le sequencing and a MiSeq
for sample evaluation or library testing.

Stem Cell and Xenograft Core:
The Stem Cell and Xenograft Core (SCXC) is a
comprehensive resource laboratory that integrates a viable tissue bank of normal human
hematopoietic cells and hematopoietic malignancies with a full range of xenograft
services.

Study Design and Biostatistics (SDAB):
The Study Design and Biostatistics (SDAB)
Core works closely with existing resources to provide targeted study
design and
biostatistics support to ITMAT/CTSA investigators. The Core serves as a direct provider
of services, including protocol review, study design, proposal development, and
performance of simple to potentially substantial complex analyses.
