Invisible to the naked eye, the micro
-
organisms are essential to life on earth, to food
production and human life.

For the first time, a Moroccan focal point is created where microbiological material is
preserved and related information and data are compil
ed.

The Moroccan Co
-
ordinated Collections of Micro
-
organisms (
CCMM
) is a national
network for science and research. It contains 10 institutions belonging to four ministries
(Ministry of Higher Education and Scientific Research, Ministry of Agriculture, M
inistry of
Health and Ministry of Equipment). The network is co
-
ordinated by a central unit localized at
the “Centre National pour la Recherche Scientifique et Technique (
CNRST
)”.

The setting up of the CCMM network in Morocco resulted from a collaboration
with the
Belgian Co
-
ordinated Collections of Micro
-
organisms (BCCM) consortium which is financed
and co
-
ordinated by Belgian Office for Scientific, Technical and Cultural Affairs.

The main goal of the CCMM network is to create a national tool for promotin
g scientific
interest and investigations in microbial biodiversity within the Moroccan academic and
bioindustry communities. It is responsible for the distribution to interested academic and
industrial workers
.

The CCMM network takes into account th
e Convention on Biological Diversity (CBD, Rio
de Janeiro, 5 June 1992). The CBD aims to provide for «

the conservation of biological
diversity, the sustainable use of its components and the fair and equitable sharing of the
benefits arising out of the uti
lization of genetic resources

» (CBD article 1). At present, 160
countries including Belgium and Morocco, have signed this convention.


The present research activities of the CCMM network are financed mainly by the Ministry
of Higher Education and Scienti
fic Research, the National Center for Scientific Research
(CNRST), and Belgian General Administration for International Cooperation (DGCI).

The CCMM collections compile approximately 3000 strains of micro
-
organisms including
bacteria, yeasts, fungi and alg
ae. For its first edition of the catalogue, the CCMM publish 447
well characterized and documented strains (bacteria, yeasts and fungi) of Moroccan origin.

The CCMM culture collection contains 447 strains of bacteria, fungi and yeasts
representing 127 species, subspecies or pathovars divided into 49 species of bacteria and
78 species of fungi and yeasts.

The coll
ection is reflected in its extensive holding of cultures employed in fermented

PRESERVATION


Longtime preservation is gu
aranteed at CCMM:



all bacterial strains are lyophilised and cryopreserved at

80°C;



most of the yeasts and fungi are lyophilised except for some strains preserved under
mineral oil (MO) and under sterile water (WA).


At CCMM, all strains are availabl
e under agar and may be delivered either at lyophilised
state or grown on an appropriate culture medium

2.1. ALPHABETICAL LIST OF STRAINS

Strains are listed alphabetically, according to their scientific name and under their
accession number in the collection.


2.2. NOMENCLATURE


Bacterial strains are generally listed under their most recent name. Names are valid only
when listed in the ‘
Approved Lists of Bacterial Names’ (indicated in the catalogue by AL) or
when validly published since then in the International Journal of Systematic Bacteriology,
either by original publication (indicated by VP) or by quotation in the validation lists (in
dicated
by VL). All other names are considered invalid.


Many species of fungi are known as pleomorphic, having two or more distinct
reproductive states or forms of reproduction. Pleomorphic species therefore have several
spore
-
states in the life cycle, th
e sexual forms the
teleomorph (“teleom”)
.The asexual form
represents one
anamorph (“anam”)
. Each state may also have synonyms.

Species are normally listed under their anamorphic name and cross
-
referenced with the
teleomorphic name. However, when the teleom
orphic state has been observed, the strain is
listed under the teleomorphic stage with cross
-
reference to its anamorphic state.

THE CCMM DATABASE


The format for computer storage and retrieval of strain
data is done following the
general structure and adopted data fields in agreement with the

icrobial

urope (MINE). This structure has been improved by François Guissart from
BCCM.

Data input is developed by Hassan Bouhaddou in CCMM.

4.1.

ISOLATION AND PURIFICATION OF MICRO
-
ORGANISMS

The C

CMM network staff can provide assistance in the purification of
cultures
, the
isolation of strains of micro
-
organisms (bacteria, yeasts or fungi) from a diverse range of
contaminated sources, the isolation of strains with specific characteristics, etc.


4.2.

CHARACTERISATION AND IDENTIFICATION OF STRAINS

A comprehensive and reliable
identification
and
characterisation

service for micro
-
organisms is available at CCMM network, using an extensive array of
phenotypic tests
(such as biochemical reactions, cell
ular protein electrophoretic patterns),
genotypic
analyses
(such as % G+C determinations, restriction fragment analysis of amplified rDNA,
hybridisation of DNA with reference DNA's, PCR analysis, etc.


4.3.

MOLECULAR BIOLOGY SERVICE


CCMM network can prov
ide at least molecular typing or fingerprinting of cultures such
as PFGE, RAPD, AFLP, rep PCR.


4.4.

STORAGE PRESERVATION AND DISTRIBUTION OF STRAINS


Compared to similar advanced collections, the CCMM network is now using suitable
techniques for preserving micro
-
organisms such as cryopreservation and lyophilisation. The
strain distribution is performed following the known criteria of quality control.


4.5.

ADVICE AND CONSULTANCY


The CCMM network can be contacted for advice and consultancy on basic
microbiology. All advice and information are provided on a confidential and impartial basis
and on a range of scientific issues of microbiological importance
.


4.6.

TRAINING

Training can be offered to introduce different approaches to strain maintenance,
Characterisation, identification and collection management. The activities range from courses
and workshops to hosting research fellows and personalised train
ing.


4.7.

EDUCATIONAL SERVICES

For teaching purposes, a set of strains has been selected which can be used to
demonstrate most of the biochemical characteristics used for common bacterial identification.


4.8.

CONTRACT RESEARCH


Research activities can be linked to projects with public funding, to international
cooperation and with industry.

IMPORT REGULATIONS

The guidelines and regulations for import of materials vary. Most count
ries have their own
import rules. Clients should therefore verify whether national and international regulations
allow them to receive the material ordered. Appropriate certificates, import licences or
mailing tags for material requiring an import permit s
hould be provided by the client.

6.1.

Revival of freeze
-
dried cultures


The CCMM freeze
-
dried cultures are supplied in vacuum
-
sealed glass ampoules on
request. Ampoules may be opened in a ordinary transfer cabinet.



Make a small file mark on the ampoule near the middle of the cotton wool plug. Touch the file
mark with a red hot glass rod till the glass cracks.

Wait a few seconds to allow air to enter into the ampoule. Remove
the upper part of the
ampoule and the cotton plug.

Add approximately 0,5 ml of appropriate broth with a sterile Pasteur pipette to the dried
material after flaming the open end of the ampoule.




Mix the contents gently with the tip of the Pasteur pipette, and transfer the contents to one or
more suitable solid and/or liquid media (follow the medium indications given in the list of
strains).

Allow the cells to rehydrate slowly on solid media. It is

advisable to include a general medium
in order to detect possible airborne contaminants which may have been introduced during
the opening of the ampoule.

All ampoule remains should be sterilized before discarding.


6.2.

Hazardous strains at CCMM


The huma
n pathogenic species of the CCMM collection are listed below, according to
the latest publications and cases of new pathogenic species.

Hazard group 1 (H1): organism that is unlikely to cause human disease in healthy person.

Hazard group 2 (H2): organism
that may cause human disease and which may be a hazard
to laboratory workers but is unlikely to spread in the community. Laboratory exposure rarely
produces infection and effective prophylaxis or effective treatment is usually available.

Hazard group 3 (H3
): organism that may cause severe human disease and present a serious
hazard to laboratory workers. It may present a risk of spreading in the community but there is
usually effective prophylaxis or treatment available.

Hazard group 4 (H4): consists of orga
nisms that cause severe human disease and are a
serious hazard to laboratory workers. They may present a high risk of spread in the
community and there is usually no effective prophylaxis or treatment. Should be handled
under containment level 4. Distribut
ion only through official permits.
