During my M.Sc. dissertation I gained little bit of expertise with tools of genetic transformatio

using tobacco as model plant. The common methods of genetic transformation viz.
Agrobacterium
mediated, biolistic and electroporation were used to transform tobacco with gene construct having
GUS reporter gene. The regeneration protocols along with isola

Triticin, a minor seed storage protein of wheat has been reported to have lysine repeats in the
hypervariable region, which can be utilized for improving the nutritional quality using
biotechnological approach since i
t represents only 5% of the total seed storage protein of wheat. The
different approaches for genetic manipulation of triticin gene can be i) increasing the copy number
ii)screening of different wheat progenitors for the possibility of better lysine repea
ts in the
hypervariable region and iii) enhancing the expression of triticin gene using strong seed storage
protein promoters. An attempt was made to clone the full
-
length cDNA of triticin gene. Seed storage
protein promoters of HMW, LMW glutenin, gliadins
, oat12S globulin, and wheat triticin were PCR
amplified and cloned in promoterless vector.
Moreover attempt was made to analyze the inherent
variation in the hypervariable region of triticin gene among different wheat progenitors based on its
PCR amplific
ation pattern.

Polypeptides and proteins that are inhibitors of proteolytic enzymes are ubiquitous in nature
and found in cells of almost all life forms. Being highly specific in their interaction with proteases
they abolish their proteolytic activity by bindin
g very tightly. Protease inhibitors belong to the seed
storage proteins and therefore they are particularly abundant in seeds and storage organs where they
accumulate to around 1
-
10% of total protein content or in exceptional cases to much higher level.
Mo
st of the protease inhibitors that have been characterized from plants do not inhibit endogenous
plant proteases but have specificities for animals or microbial enzymes. This led to the view that they
might be involved in the protection of these vulnerable

plant tissues from pest and pathogen attack by
virtue of an antinutritional interaction with the pest’s digestive enzymes.


Till now most of the efforts have been made to study PIs in cultivated varieties and not much
work has been carried out in wil
d legumes. Isolation, purification and characterization of proteinase
inhibitors from wild legumes like
Cassia fistula, Rhynchosia
and

Atylosia

has been

A grain legume of major importance to developing countries includes peanuts (
Arachis
hypogaea
), common beans (
Phaseolus vulgaris
), chickpea (
Cicer arietinum
), Cow
peas (
Vigna
unguiculata
), pigeonpea (
Cajanus cajan
) and lentils (
Lens culinaris
) etc. Pigeonpea is one of the
important high
-
protein grain legumes of semi
-
arid tropics and sub
-
tropics, particularly India
contributing to 80% of total world production. This

crop of strategic importance is highly susceptible
to number of abiotic and biotic stresses affecting the total yield. The conventational breeding methods
have not been successful in raising stress
-
resistant genotypes because of limited genetic variabilit
y
amongst cultivated accessions and sexual incompatibility with wild relatives. There is an urgent need
to widen the genetic base in pigeonpea and incorporate desirable traits using transgenic technology.



Efficient protocols for successful transformatio
n are quite limited in pigeonpea due to its
recalcitrance to regeneration in tissue culture, which is the prerequisite to any successful transgenic
productions.
In vitro

regeneration of pigeonpea has been reported earlier and has been recently been
followe
d by many researchers, and still there is paucity of an efficient regeneration protocol that could
be effective to agronomically important Indian cultivars. Moreover there is wide dominance of the
genotypic factor on
in vitro

regeneration of complete plant
lets, and the reported regeneration
protocols are limited to region specific cultivars. With this background few agronomically important
local cultivars of pigeonpea having maximum acreage of cultivation in the north
-
eastern terrain region
adjoining Gorak
hpur district is being screened for in vitro regeneration ability. Initial standardization
of direct regeneration protocol in one of the cultivar of pigeonpea, ND
-
1 from Faizabad has been done
with some encouraging results. Presently screening is in progre
ss for 15 different cultivars. The
objective of the study being to standardize a reproducible protocol for the direct shoot organogenesis
using potential explants from
in vitro

grown pigeonpea that could be amenable to the different
methods of gene transfe

and substantial loss of 12 species of
Aspergillus

A. carneus, A. deflectus, A.
melleus, A. panamensis, A. petrakii, A. puniceus, A. quardilineatus, A. regulosus, A. sparsus, A.
subolivac
eus, A. sulphureus and A. terricola.
Four species of
Aspergillus

namely

A. flavo
-
fercatis, A.
oryzae, A. tubingensis
and

A. zonatus

were reported in the present study which were not observed in
earlier study. Moreover diversity of
Aspergillus

in terms of

number of strains of different species
revealed
A. flavus

to be the dominant species as compared to
A. niger

mentioned in earlier report
isolated from soil of kusmi forest.The shift in the diversity of
Aspergillus

is directly related with the
shift in the

vegetation of study site over the time. The present teak dominated vegetation has
experienced a loss of 15 tree species and 37 annuals while there is inclusion of 22 new annuals when
compared with earlier report.

A total of 28 indigenously isolated fungal

strains for amylase and protease secretion have

As Principal Investigator in one of the project in DBT funded Programme mode Support in
Agricultural Biotechnology entitled “
Investigating the rol
e of Dof transcription factor in
accumulation of seed storage proteins and nitrogen metabolism during grain development in finger
millet”


As per the objectives of the project an attempt has been made to design primers specifically
for Dof domain/ genes ba
sed on sequences published in literature for different Dof genes of cereals
viz
. rice, wheat, barley, maize to be utilized for PCR amplification of probable Dof domain/ genes
from finger millet. The major outcome has been the amplification of Dof domain fr
om finger millet
using primers designed from rice sequences, which has further been verified by sequencing the
amplicons. Attempt has been made to amplify putative dof genes (Full
-
length/ Partial) from finger
millet using primers designed from other cereal
s. The sequenced amplicons representing putative Dof
genes from rice, maize, barley and finger millet has been confirmed by
in silico

analysis.

Further an attempt was made to study the phylogenetic relationships among cereals
(rice,
wheat, barley, maize, o
at, sorghum)
and millets

(finger millet, barnyard millet, proso millet, little
millet, kodo millet, foxtail millet)
based on the PCR amplification pattern using dof domain related
primers. The domain specific primers resulted in multiple band formation wit
h variability among
cereals and millets in terms of both number of bands and sizes. The cloning and sequencing of all the
bands might give certain insight into the largely unknown genome of finger millet through
comparative genomics. This may have some imp
lication regarding their syntenic relationships
provided that all are sequenced and mapped onto respective genomes. The DNA polymorphism
observed might suggest the potential use of gene sequences as functional markers for diversity study
of agronomically i
mportant cereal crops.

Pectin lyase being one of the important enzymes of
group pectinases is relatively less
explored compared to others like polygalactuornases and pectate lyases. Isolation, purification
and application of pectin lyases produced from indigenous isolates of
Aspergillus

Penicillium

citrinum

has been carried
out. The pectin lyases produced from these sources were purified,
characterized and their possible industrial application in fruit juice clarification and retting of
natural fibers has been studied. Prior to cloning of microbial pectin lyase gene(s), the
