Genetic Engineering: Course description
The course is organized in lectures and laboratory practice and its aim is giving to students basic knowledge
for constraction and use of cloning and expression vectors containing recombinant DNA in prokaryotic and
eukaryotic system. Special attention will be directed to study biological systems utilized for the large scale
production of recombinant autologous or eterologous proteins, focusing on advantages and disadvantages of
each system, to allows students to evaluate and solve problems related to the expression of recombinant
proteins. Furthermore, during the course, the major applications of genetic engineering in biotechnology will
be presented, to let the student undertand the power and the potential hazards of the acquaired technical
skills. The laboratory practice will be organized as short research program.
Objectives of the course
After the course the student should: (a) have acquaired the theorical and pratical skills necessary to constract
and use recombinant DNA molecules and vectors, for the isolation characterization and expression of genes;
(b) know the most common vectors used for cloning and producing recombinant proteins; (c) decide,
according to different use/source of recombinant proteins, which biological system is optimal for expression;
(d) evaluate the advantges and risks in the use of genetic engineering in different biotechnological fields.
Genetic Engineering: Prerequisites and
Basic knowledge of DNA structure and functions, protein sctructures and functions, and good knowledge on
prokaryotic and eukaryotic cells.
Genetic Engineering: Course contents and
-Basic Molecular Biotechnology
- Prokaryotic and Eukaryotic Host systems; restriction enzymes and other
enzymes usefull for DNA and RNA manipulations; chemical synthesis, sequencing and amplification of DNA;
site directed mutagenesis; search gene in gene banks and computer analysis of data. Cloning and
expression vectors based on plasmid and bacteriophage; transfection and selection methods; constraction of
genomic and cDNA libraries; large scale production of recombinant proteins.
- Expression vector; transfection and selection methods utilized in yeast, insect cells and mammalian cells;
production of recombinant protein in eukaryotes; vectors for gene therapy.
Application in Biotechnology
- production of therapeutic agents; synthesis of commercial products througth
recombinant microrganisms; gene therapy and transgenic animals.
