In a previous research project carried out by this group at DISTA, University of Bologna

we
carried out a QTL study for stress
-
adaptive traits and yield across
multiple

field trials in the
Mediterranean basin exploring a broad range of soil moistures and temperatures. This
study investigated a panel of 189
elite
cultivars suitable for associa
tion
mapping
(Maccaferri et al. 2011
) and a biparental RIL population (Maccaferri et al. 2008). A major
QTL for grain yield per se on chr. 3BS (Qyld3BS) was shown to consistently affect yield
across environments. Pairs of near isogenic lines (NILs) for Qyl
d3BS have been produced
and the positional cloning of Qyld3BS is well underway at UNIBO. These NILs together
with the panel of 189 accessions are ideal for testing the genetic basis of heat tolerance on
yield and yield

stability.

TRAINING WORK PLAN


Objective

The aim is to train a
post
-
doc

in the molecular
genetics and
breeding in cereals in general
and in particular in

durum wheat
. The
post
-
doc

will acquire experience and skillness in the
various aspects

that are necessary to stud
y the genetic control of important agronomic
traits in durum wheat, both on the molecular biology and field experiment sides.


Research activities


The candidate will be involved in the

following main actions:


1.

Association mapping (AM) to identify QT
Ls that control heat tolerance.
A collection
of 189 elite cultivars

(Maccaferri et al. 2011)

will be evaluated
in both open
-
field and
greenhouse trials
.
For the phenotypic evaluation, i
n addition to grain yield and its
components,

other yield
-
related trait
s will be considered.

2.

Evaluation of pairs of near isogenic lines (NILs) for Qyld3BS

in different locations
and planting dates
.

3.

Evaluation of heat and drought tolerance of the parental lines of
different

mapping
populations

in order to identify a valuable

resource to carry out a biparental
mapping experiment.

4.

Characterization of plant materials derived from crosses involving durum wheat and
the wild grass Thinopyrum ponticum (T. ponticum is a donor of chromosome
segments influencing grain yield).



Requir
ements

Ph.D in plant
genetics, agronomy,
molecular biology
, plant biotechnology, plant breeding

or other related disciplines with experience in plant
(especially cereal crops)
physiology
and genomics and good molecular biology skills. It is required experience in DNA isolation,
genotyping,
QTL analyses,

bioinformatics and development of sequence
-
based markers.
Proven ability to manage experimental activities (lab, greenhouse and

field) required for
the collection of phenotypic data will also be positively evaluated. A good knowledge of
written and spoken English is required.


Location of research activities.

The research activities will be carried out in the laboratories of the
Department of
Agr
iculture (DIPSA)
, University of Bologna and in the Experimental Station of the same
University.
The candidate will interact with other researchers in cereal genetics and
genomics for some aspects of the phenotyping, genetic stocks developm
ent and
genomics
-
related activities.
